In recent years, the use of nonsteroidal anti-estrogen (NSTA) in bodybuilding has been recognized as a significant advancement in the treatment of gynecomastia (). This was particularly concerning because of the remarkable response of the majority of men to NSTA. The development of the NSTA class of compounds, such as tamoxifen and clomiphene, was first synthesized in the late 1970s and was subsequently used for the treatment of gynecomastia (). These compounds were subsequently shown to be highly selective for the aromatase enzyme (). Moreover, they are widely used in the treatment of gynecomastia because of their ability to block the conversion of estrogen into the active metabolite 5α-reductase (). In addition to the estrogen-receptor blockade, NSTA has also been shown to be a potential target for antiestrogen treatment for gynecomastia ().
NSTA has emerged as a potential drug class for the treatment of gynecomastia because of its ability to block the conversion of estrogen into the active metabolite 5α-reductase (). It is well established that this enzyme is a crucial enzyme involved in the conversion of estrogen into the active metabolite 5α-reductase. Moreover, it has also been shown that the effects of NSTA on the estrogen-dependent pathways may be dependent on the presence of estrogens, as well as the presence of nonestrogenic estrogens such as a progestogen (). In contrast, studies have shown that NSTA may act in the opposite direction to estrogen in the opposite tissue, with effects on the estrogen-dependent pathways being greater in the opposite tissue than in the estrogen-independent pathways (). This may explain the differences between NSTA and estrogen that may be observed in the development of the various antiestrogens. The main mechanism by which NSTA functions in the development of gynecomastia is based on its ability to block the conversion of estrogen to the active metabolite 5α-reductase (). This has also been the focus of many studies that focused on the effects of NSTA on the estrogen-dependent pathways. In addition, it has been demonstrated that the effects of NSTA on the estrogen-independent pathways may be dependent on the presence of estrogens, as well as the presence of nonestrogenic estrogens such as a progestogen (). These studies have been carried out in different tissues such as breast, cervical, and oral tissues ().
In this study, we investigated the effects of NSTA on the estrogen-dependent pathways in breast, cervical, and oral tissues of menopausal women.
Nolvadex® (Nolvadex®; Novartis, Basel, Switzerland) was purchased from Sigma-Aldrich (Steinheim, Germany) and was used within a minimum of five days. Nolvadex® was dissolved in deionized water and added to a final concentration of 0.05 mg/ml. Nolvadex® was dissolved in deionized water and was then added to a final concentration of 0.15 mg/ml.
